Abstract
WE have found that the reactions citrate ⇌ cis-aconitate (i) isocitrate ⇌ cis aconitate (ii) can be separated during a controlled ageing at 0° C of aconitase (aconitate hydratase, E.C. 4.2.1.3.), purified as far as the first ethanol stage in Morrison's preparation from pig heart. At this stage, the enzyme still contains the Fe2+ combined with the enzyme, so that no activation is necessary1. In Fig. 1, curves (a) and (b) give the initial rates for (ii) and (i) respectively, ratio (i)/(ii) 1.00. After 6 days at 0° C, the rates observed in the same conditions were (c) and (d), making the ratio 0.18.
