Advances in chimeric antigen receptor (CAR)-T cell therapies have revolutionized the treatment of many haematological cancers. However, for solid tumours, targets must be identified and the accumulation, persistence and function of the CAR-T cells must be improved. Two recent advances aim to address these issues. In Science, vaccination with RNA encoding claudin 6 (CLDN6), combined with a CAR-T cell therapy that recognized CLDN6, regressed large tumours in mouse models. In a paper from another group in Nature, altering the metabolism of CAR-T cells by deleting REGNASE-1 rendered these cells long-lasting and able to combat melanoma and acute lymphocytic leukaemia (ALL) in mice.
