Enzymatic Dehydrogenation of Hydroxypurines and Hydroxypteridines

Abstract

ONE of the steps of the biosynthesis of guanylic acid from adenylic acid involves the diphosphopyridine nucleotide-linked dehydrogenation of inosinic acid to xanthylic acid1–3: inosinic acid+diphosphopyridine nueleotide+ → xanthylic acid + reduced diphosphopyridine nucleotide. When this reaction was studied with chicken-liver enzyme extracts by following the appearance of the absorption peak at 340 mµ characteristic of reduced diphosphopyridine nucleotide, a strong reduction of the coenzyme was observed not only in the presence of inosinic acid but also with the corresponding purine base hypoxanthine.
